Abstract


Pewarnaan bakteri merupakan salah satu materi dari Praktikum Mikrobiologi yang harus ditempuh oleh mahasiswa di salah satu Program Studi Vokasi Kesehatan. Penggunaan pewarna bakteri sintetis berbahaya terhadap manusia dan lingkungan hidup karena bersifat mutagenik. Penelitian ini bertujuan untuk mengetahui potensi dari ekstrak kelopak bunga telang (Clitorea ternatea L.) sebagai alternatif pewarna bakteri sederhana berbasis bahan alam ramah lingkungan. Metode penelitian yang digunakan adalah eksperimen laboratorium dengan menggunakan analisis secara deskriptif. Hasil penelitian menunjukkan ekstrak kelopak bunga telang (C. ternatea L.) memiliki kemampuan untuk mewarnai sel bakteri Gram-positif Staphylococcus aureus dan bakteri Gram-negatif Escherichia coli dengan kontras yang dapat dilihat di mikroskop. Bila dilihat dari kualitas pewarnaan nya, preprat yang diwarnai dengan ekstrak kelopak bunga telang memberikan kualitas pewarnaan yang kurang baik bila dibandingkan pewarna bakteri seperti safranin dan kristal violet. Kesimpulan dalam penelitian ini adalah ekstrak kelopak bunga telang (C. ternatea L.) belum bisa dipergunakan secara langsung sebagai alternatif pewarna bakteri sederhana berbasis bahan alam ramah lingkungan karena membutuhkan penambahan co-pigment sebagai stabilisator dari antosianin.
