Abstract
Obturation is the last step in root canal treatment to prevent recolonization of bacteria remaining in the root canal and prevent the invasion of bacteria from the outside of the root canal, by closing the entire access that connects between periodontal and root canal. Selection of root canal sealer can support to the successfull of root canal treatment. Enterococcus faecalis is a bacteria
that causing the failure of root canal treatment, because these bacteria can survive in the root canal that has been obturated.
This study aims to determine the antibacterial activity of root canal sealer against E. faecalis. This study investigated the antibacterial activity of root canal sealer by using diffusion technique. E. faecalis with a density of 9 x 108 CFU/ml were inoculated in Mueller Hinton agar. Three wells made on agar medium with a diameter of 6 mm and a depth of 4mm at the same length in a petri dish measuring 10 mm x 100 mm. Each well at the bottom of the dish was marked by a marker to indicate a type of sealer inserted into the wells. Code A was a group of zinc oxide eugenol-based sealer, code B was a group of epoxy resin-based sealer and code C was a group of mineral trioxide aggregate-based sealer. Approximately 0.1 ml of
each sealer was taken, included into the well by using plastic instrument and split injection of 1 ml. The plates were re-incubated at 37°C for 24 hours, then zones of inhibition were measured. The data obtained were subsequently recorded and analyzed by using the one-way ANOVA and the Least Significant Difference (LSD) test.
ANOVA results indicate there are differences antibacterial activity by sealer based of zinc oxide eugenol, epoxy resin and mineral trioxide aggregate against E. faecalis (p <0.05). In the LSD test (p <0.05), the mean difference between the groups
shows that sealer based of zinc oxide eugenol has an antibacterial activity better than epoxy resin and mineral trioxide aggregate against E. faecalis.
