Abstract
Background. Pathological condition in diabetes mellitus patients may associate with several complications, including bone-healing disorder. Simvastatinis knownascholesterol-lowering drugs whichhave been widelyusedtoincrease bone massby inducingbone morphogeneticprotein-2 (BMP-2) on osteoblastsandstimulatebone formation. Local application of simvastatin
will induce and accelerate bone formation locally and trigger early expression of growth factors that regulates angiogenesis, differentiation of bone cells and osteogenesis. Type I collagen is one marker of bone formation in the healing process of bone produced by osteoblasts. This study aims to describe the effect of application simvastatin in the process of bone healing to type I
collagen expression on rats model with type 1 diabetes mellitus.
Methods. Experimental research with 45 Sprague Dawlay rats was divided to 3 groups. Group 1 is 15 normal rats with defect in the left femurs. They were given with simvastatin gel and the right femurs defects were left without it. Group 2 is 15 DM
rats with defects in the left femurs. They were given with CMC-Na gel, while the right femurs were not. Group 3 is 15 DM rats with defects in the right femurs and filled with simvastatin gel. All groups were decapitated on day 5th, 8th and 10that post manufacture defect. Finallyan immunohistochemistry (IHC) examination conducted in all samples to examine the expression of type I collagen
according to the observations days.
Results. Total percentage of type I collagen expression between diabetic rats that were given simvastatin gel compared to DM rats which were not given with simvastatin gel resulted significant difference (P <0.05). Conclusion. Application of simvastatin on bone rats model with type 1 diabetes mellitus significantly increased the percentage of type 1 collagen expression during the bone healing process.
