Blood Sugar-Lowering Activity and Acute Toxicity of Eleutherine bulbosa Bulbs Extract from Lampo Donggala, Central Sulawesi, Indonesia

Abstract
Eleutherine bulbosa is a medicinal plant with pharmacological properties, including antidiabetic properties. Therefore, this study aims to determine the effect of administering 150 mg/kg body weight (BW) of crude and purified extract of E. bulbosa bulbs from Lampo, Central Sulawesi, Indonesia, on blood glucose reduction in streptozotocin-induced rats, and the acute toxicity of the extract. The maceration method was used to perform extraction with 50% methanol to obtain the crude extract. Subsequently, the purified extract was derived through column chromatography and liquid-liquid partition methods. Blood glucose levels were measured before induction (H0), 3 days post-induction (H3), and 28 days post-extract administration (H31) using a glucometer. A total of 20 male rats, divided into 5 groups, were observed for symptoms of acute toxicity in 24 hours, and the number of dead subjects was recorded. Meanwhile, the percentage reduction in blood glucose levels in the negative control, positive control, water column extract, purified extract, and crude extract groups was 0%, 82.1%, 66.8%, 72.7%, and 69.3%, respectively. The purified extract was not significantly different from glibenclamide as a positive control, with p<0.05. The acute toxicity test of the purified extract of E. bulbosa showed that there was no record of deaths. The LD50 was approximately 2000 mg/kgBW, representing a criterion of practical non-toxicity.
