THE EFFECT OF MEDIUM ON ENDOPHYTIC FUNGUS IP-2 GROWTH AND PRODUCTION OF ITS ACTIVE INHIBITOR HEME POLYMERIZATION METABOLITE

Abstract
Endophytic fungus IP-2 is the endophytic fungus isolated from the plant Artemisia annua L. This fungus is known to have activity as an inhibitor of heme polymerization. The aim of this study is to examine the suitable medium for the growth and production of the active metabolite and to determine the appropriate time for harvesting the active metabolite. Endophytic fungus was fermented in 4 types of media for 14 days. Sampling was taken every day to obtain the weight of the dry cells which were used for analysis of growth. The media obtained is used to determine their activities as heme polymerization inhibitor. Tests were carried out using the method developed by Bassilico et al (1998). Data of dry cell weight and the percentage inhibition of heme polymerization were analyzed in order to obtain the curve of growth and metabolite production. Both curves will be used to determine the proper time to harvest the active metabolite. The results showed that the PDB was the best medium for endophytic fungus IP-2 growth and was able to produce the active metabolite higher than most other media. In the PDB media, at day-7 showed that active metabolite produced at most so that proper harvest time metabolite is on the 7th day of fermentation.
